Background

TAK1 (MAP3K7) is a ubiquitously expressed serine/threonine kinase that acts as a central signaling hub by integrating cues from TGF-β, inflammatory cytokines (TNF-α, IL-1), pattern-recognition receptors (TLR, NOD-like receptors) and cellular stress to catalyze the activation of IKK and MAP2Ks (MKK3/6, MKK4/7), thereby driving the NF-κB, p38, JNK and ERK pathways that collectively regulate inflammation, innate and adaptive immunity, cell survival, differentiation and metabolic adaptation; its activity is tightly controlled through stimulus-inducible assembly with regulatory binding partners TAB1, TAB2 and TAB3 and by polyubiquitin scaffolds, and genetic or pharmacologic dysregulation of TAK1 is linked to inflammatory diseases, autoimmunity and cancer.
