Background
SERPINA10 encodes the 72-kDa plasma serpin known as protein Z-dependent protease inhibitor (ZPI), a 444-aa liver-secreted glycoprotein that exerts potent anticoagulant activity by selectively inhibiting membrane-associated factor Xa (in a reaction that strictly requires its vitamin-K-dependent cofactor protein Z, Ca²⁺ and anionic phospholipids) and factor XIa (even in the absence of cofactors); mechanistically, ZPI presents a unique tyrosine-centered reactive-center loop that docks into the protease active site, forming a tight inhibitory complex whose kinetics are accelerated by heparin but can be antagonized by oxidative lipid modifications, while structural studies reveal a canonical serpin fold with specialized protein-Z-binding determinants that allosterically position ZPI on pro-coagulant membranes, and clinically SERPINA10 deficiency or missense mutations are linked to increased risk of venous thrombosis, recurrent miscarriage, and altered expression is observed in cancers and pregnancy-related disorders .
