IHC shows positive staining in paraffin-embedded human stomach. Anti-Pericentrin antibody was used at 1/2000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Background
Pericentrin (PCNT, also called kendrin or pericentrin-B) is a large, highly conserved coiled-coil scaffold protein encoded by the PCNT gene on chromosome 21, which localizes to the centrosome and pericentriolar material (PCM) via its C-terminal PACT domain, anchoring γ-tubulin complexes and other regulatory proteins to organize microtubule nucleation and anchoring, thereby orchestrating centrosome assembly, mitotic spindle formation, cell-cycle progression and primary cilium biogenesis; mutations or dysregulation of PCNT underlie disorders such as microcephalic osteodysplastic primordial dwarfism type II, Seckel syndrome, ciliopathies, and have been linked to cancer, mental disorders and altered insulin secretion in β-cells.

Immunocytochemistry
ICC shows positive staining in HeLa cells. Anti-Pericentrin antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
