Background
IL1RAP (interleukin-1 receptor accessory protein) is a transmembrane glycoprotein that serves as the indispensable co-receptor for all members of the IL-1 cytokine family (IL-1α, IL-1β, IL-33, IL-36) by forming a trimeric ligand–primary receptor–IL1RAP complex whose intracellular TIR domains recruit MyD88, triggering sequential activation of IRAK4/IRAK1/2, TRAF6 and downstream MAPK (p38, JNK, ERK) and NF-κB pathways that drive inflammation, cell survival and proliferation; physiologically expressed at low levels in most normal tissues, IL1RAP is markedly up-regulated on the surface of hematologic and solid tumor cells where it additionally interacts with and amplifies oncogenic receptor tyrosine kinases such as FLT3 and c-Kit, and its high expression correlates with poor prognosis, making it a promising selective target for antibody, CAR-T and small-molecule therapies.

Immunocytochemistry
ICC shows positive staining in A673 cells (top panel) and negative staining in raji cells (below panel). Anti-IL1RAP antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
