Background
Histone H3 tri-methylated lysine 9 (H3K9me3) is a crucial epigenetic mark formed by the trimethylation of the ninth lysine residue on the core histone H3. It is widely recognized as a classic hallmark of constitutive heterochromatin. Its primary function is to recruit effector proteins like Heterochromatin Protein 1 (HP1), which subsequently promotes the compaction of chromatin into a dense, transcriptionally silent higher-order structure. This modification plays a key role in maintaining genomic stability by effectively repressing the aberrant expression of repetitive genomic elements, such as transposons and telomeric regions, as well as foreign genes. Furthermore, H3K9me3 is essential for the transcriptional regulation of specific genes, X-chromosome inactivation, and cell fate determination. It is important to note that aberrant distribution of H3K9me3 is closely associated with various diseases, including cancer development, neurodegenerative disorders, and the aging process.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human colon. Anti-Histone H3 (tri methyl K9) antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

ICC shows positive staining in HeLa cells. Anti-Histone H3 (tri methyl K9) antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
