Background
ER alpha (estrogen receptor alpha) is a 66-kDa ligand-activated transcription factor belonging to the nuclear receptor superfamily that, upon binding 17β-estradiol, translocates to the nucleus, homodimerizes, and recruits co-regulators to estrogen response elements (EREs) in target genes, thereby governing proliferation, differentiation, and survival in reproductive tissues, bone, cardiovascular system, and CNS, while its dysregulation drives breast and endometrial cancers, making it the principal therapeutic target of selective modulators (SERMs) and degraders (SERDs).

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry
ICC shows positive staining in T-47D cells (top panel) and negative staining in SK-BR-3 cells (below panel). Anti-ER alpha antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
