Background
Cardiac troponin T (TNNT2) is a 35.9 kDa, 295-amino-acid striated-muscle-specific protein that forms the troponin T component of the troponin complex on the thin filament, where it tethers the troponin C–I sub-complex to tropomyosin; by transducing the Ca²⁺-induced conformational change of troponin C into tropomyosin movement, TNNT2 orchestrates the steric switch that exposes myosin-binding sites on actin and thus initiates systolic contraction, while its unique N-terminal peptide sequence allows ultra-sensitive immunoassay detection—making any elevation in blood TNNT2 the gold-standard biomarker for myocardial injury—yet mutations that enhance or suppress myofilament Ca²⁺ sensitivity, destabilize the N-tail, or promote aberrant splice variants are linked to hypertrophic, dilated, or restrictive cardiomyopathies, arrhythmogenic right ventricular cardiomyopathy, and sudden cardiac death, underscoring its dual role as both a molecular motor controller and a sentinel of cardiac integrity.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human cardiac muscle. Anti-Cardiac troponin T (TNNT2) antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
Negative control: IHC shows negative staining in paraffin-embedded human skeletal muscle. Anti-Cardiac troponin T (TNNT2) antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin.
