Flow cytometric analysis of Human Granzyme B expression in Human PBMC. Human PBMC were fixed with 4% PFA and permeabilized with 0.1% Tween. The cells were stained with Alexa Fluor® 647 Mouse Anti-Human CD3 Antibody and either Alexa Fluor®488 Mouse IgG1, κ Isotype Control (Left panel) or Alexa Fluor®488 Mouse Anti-Human/Mouse Granzyme B Antibody (Right panel) at 5 μl/test. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
Granzyme B is a 32~kDa serine protease stored in the cytolytic granules of cytotoxic T lymphocytes and natural killer cells, whose immune function centers on perforin-assisted translocation into target cells where it directly cleaves and activates pro-apoptotic substrates such as procaspase-3, Bid and PARP, thereby igniting both caspase-dependent and caspase-independent mitochondrial apoptosis pathways to eliminate virus-infected or malignant cells.
