Background
YTHDF1 (YTH N6-methyladenosine RNA binding protein F1) is a cytoplasmic reader of the m6A epitranscriptomic mark that selectively binds N6-methylated adenosines within mRNAs and, by recruiting eIF3 and other translation initiation factors, enhances the ribosome loading and protein synthesis of its target transcripts, thereby influencing cell proliferation, synaptic plasticity, immune responses and tumorigenesis; structurally it contains an N-terminal intrinsically disordered region and a C-terminal YTH domain that confers high-affinity m6A recognition, and its expression or phosphorylation state is frequently dysregulated in multiple cancers, making it an emerging therapeutic target for small-molecule or oligonucleotide-based interventions.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-YTHDF1 antibody was used at 1/250 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunofluorescence

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
