Background
VSIG4 (V-set and immunoglobulin domain-containing 4), also known as CRIg, is a transmembrane glycoprotein of the B7-related family that is selectively expressed on resting tissue macrophages; it functions both as a potent negative regulator of T-cell activation and IL-2 production and as a complement receptor that binds C3b/iC3b-opsonized particles, thereby mediating rapid phagocytic clearance of pathogens while inhibiting the alternative complement pathway convertase to limit inflammation and maintain peripheral immune tolerance.

IHC shows positive staining in paraffin-embedded human colon. Anti-VSIG4 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
