Background
Vascular Endothelial Growth Factor A (VEGF-A) is a 45 kDa homodimeric glycoprotein and the founding member of the VEGF family, encoded on chromosome 6p21.3 and expressed as at least seven splice isoforms (e.g., VEGF-A165) that are secreted or extracellular-matrix-bound to initiate angiogenesis and vascular permeability through binding VEGFR-2 on endothelial cells and neuropilin co-receptors, triggering PI3K-Akt, PLCγ-MAPK, and Src signaling cascades that drive endothelial proliferation, migration, and tube formation, while its transcription is up-regulated by hypoxia via HIF-1α and by growth factors such as PDGF and FGF, and its dysregulation underlies tumor neovascularization, diabetic retinopathy, and inflammatory disorders, making VEGF-A a pivotal therapeutic target for anti-angiogenic cancer drugs and anti-VEGF antibodies like bevacizumab.
