Background
Terminal deoxynucleotidyl transferase (TdT), also called DNA nucleotidylexotransferase or terminal transferase, is a template-independent DNA polymerase encoded by the DNTT gene on human chromosome 10q23-q24 that is selectively expressed in immature lymphoid precursors (pre-B and pre-T cells) and certain leukemias/lymphomas; it catalyzes the non-templated addition of deoxynucleotides to the 3'-OH ends of DNA, generating N-region diversity at V(D)J junctions during immunoglobulin and T-cell receptor gene rearrangement, thereby expanding antigen-receptor repertoires critical for adaptive immunity, and can utilize a broad range of divalent cations (Mg²⁺, Mn²⁺, Zn²⁺, Co²⁺) as cofactors .

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human thymus. Anti-TdT antibody was used at 1/2000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
Negative control: IHC shows negative staining in paraffin-embedded human kidney. Anti-TdT antibody was used at 1/2000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use).
