Flow cytometric analysis of FMC63 scfv Protein expression on human PBMC (human peripheral blood mononuclear cell). Human PBMC were stained with FMC63 scfv Protein (right panel) at 0.1ug/test or unlabel (left panel). APC anti-His tag antibody was used as the secondary antibody. Flow cytometry and data analysis were perfommed using Agilent NovoCyte Quanteon and FlowJorM software.

Background
FMC63 scFv is a recombinant single-chain variable fragment constructed from the VH and VL domains of murine anti-human CD19 IgG2a monoclonal antibody FMC63 linked by a flexible peptide linker; it retains high specific binding affinity to human CD19 broadly expressed on normal B cells and B-cell malignant tumors, serves as the classic antigen-recognition extracellular domain for CD19-targeted CAR-T cells adopted in four FDA-approved CAR-T therapeutics including Kymriah and Yescarta, and is recombinantly manufactured for preclinical immunotherapy construction, CD19 binding verification, CAR detection and affinity characterization assays.
