Background
NKX3.1 is a prostate-specific homeodomain transcription factor that acts as a dose-dependent tumor suppressor by integrating developmental and metabolic cues: in the nucleus it partners with the androgen receptor, FoxA1 and serum response factor to coregulate differentiation genes and a “protein-trafficking” network that includes RAB3B, while in mitochondria it is imported via HSPA9 in response to oxidative stress, binds mtDNA and up-regulates electron-transport-chain genes to restore oxidative phosphorylation, thereby limiting ROS-induced DNA damage; haploinsufficiency, allelic loss or cancer-associated point mutations (e.g., T164A) disrupt these functions, leading to prostatic intraepithelial neoplasia, progression to castration-resistant disease, and reduced survival, whereas high mitochondrial NKX3.1 predicts favorable prognosis, and secreted NKX3.1 can even act as a para-transcription factor to inhibit growth of neighboring cells, illustrating its multifaceted role in prostate development, metabolic homeostasis and cancer suppression.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry
ICC shows positive staining in LNCaP cells (top panel) and negative staining in PC-3 cells (below panel). Anti-NKX3.1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
