Background

CTLA-4 (Cytotoxic T-Lymphocyte-Associated Protein 4) is a 223-amino-acid transmembrane glycoprotein receptor expressed mainly on activated CD4+ and CD8+ T cells that functions as a critical negative regulator of immune responses by out-competing CD28 for binding to the costimulatory ligands CD80 (B7-1) and CD86 (B7-2) on antigen-presenting cells, thereby transducing inhibitory signals that attenuate T-cell proliferation, cytokine production, and cell cycle progression; the protein’s signal peptide is cleaved to yield a disulfide-linked homodimer whose extracellular Ig-like V-type domain mediates high-avidity ligand engagement, while its cytoplasmic tail contains a tyrosine-based motif that recruits phosphatases such as SHP-2 and PP2A to propagate inhibitory cascades, and genetic polymorphisms or therapeutic blockade of CTLA-4 (e.g., the monoclonal antibody ipilimumab) can enhance antitumor immunity but also precipitate autoimmune toxicities, underscoring its pivotal role in maintaining immune homeostasis.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human tonsil. Anti-CTLA4 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

IHC shows positive staining in paraffin-embedded Hodgkin’s lymphoma.

IHC shows positive staining in paraffin-embedded human gastric cancer.

Immunocytochemistry

ICC analysis of human PBMC cells treated with PHA (10µg/ml, 2 days) (top panel) and untreated human PBMC cells (below panel). Anti-CTLA4 antibody was used at 1/50 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).

ICC analysis of mouse splenocytes cells treated with Concanavalin A (2.5µg/ml, 3 days) (top panel) and untreated mouse splenocytes cells (below panel).
