Background
NRGN, or neurogranin, is a brain-specific protein found predominantly in telencephalic neurons. It is known to be a postsynaptic protein kinase substrate that binds calmodulin in the absence of calcium. NRGN is also under the control of thyroid hormone in specific neuronal subsets, playing a role in the mental states associated with hypothyroidism during development and in adult subjects. The gene is located on chromosome 11q24 and contains four exons and three introns, with exons 1 and 2 encoding the protein. NRGN is considered a direct target for thyroid hormone in the human brain, and its expression is crucial for understanding the impact of thyroid function on cognitive and mental health.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-NRGN antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
Negative control: IHC shows positive staining in paraffin-embedded human liver. Anti-NRGN antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use).
