ICC shows positive staining in HeLa cells. Anti-DNA:RNA hybrid antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Mouse IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).

Background
RNA:DNA hybrids serve as primers for DNA replication, regulate gene expression and are structural components of specific eukaryotic genomic elements, such as telomeres, centromeres and ribosomal DNA. RNA:DNA hybridization is also a crucial step in cell-specific processes, such as immunoglobulin class-switch recombination (CSR) in activated B cells and CRISPR/Cas9-mediated adaptive immunization in prokaryotic cells.

Immunocytochemistry
ICC shows positive staining in NIH/3T3 cells. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
