Background
CAD protein, also known as carbamoyl-phosphate synthetase 2, aspartate transcarbamoylase, and dihydroorotase, is a multifunctional enzyme complex that catalyzes the first three steps in the de novo synthesis of pyrimidine nucleotides. This process is crucial for the production of nucleic acids, active intermediates, and cell membranes, and plays a key role in cellular and organismal physiology. CAD is regulated by various pathways, including the mitogen-activated protein kinase (MAPK) cascade, and its dysregulation can lead to diseases such as cancer and neurological disorders.

Immunohistochemistry

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry
ICC shows positive staining in HEK293 cells. Anti- CAD/BM1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
