Flow cytometric analysis of Human CD200R expression on human peripheral blood Leukocytes. Human peripheral blood Leukocytes were stained with Brilliant Violet 421™ Mouse Anti-Human CD4 antibody and either Biotin Mouse IgG1, κ Isotype Control (Left panel) or SDT Biotin Mouse Anti- Human CD200R antibody (Right panel) at 5 μl/test followed by Sav-PE. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
CD200R is a trans-membrane glycoprotein of the immunoglobulin superfamily encoded by CD200R1, expressed chiefly on myeloid cells (macrophages, dendritic cells, microglia) and some lymphoid subsets; when engaged by its ligand CD200 (OX-2) it becomes tyrosine-phosphorylated in its cytoplasmic NPxY motif, recruits the adaptor Dok-2 and RasGAP, thereby dampening Ras/MAPK and NF-κB signaling to suppress production of TNF-α, interferons and iNOS, curb mast-cell degranulation, restrain ILC2 effector cytokines, and broadly down-regulate innate and adaptive inflammatory responses, a signaling axis that is exploited by tumors for immune evasion and is being therapeutically targeted to treat asthma, neuroinflammation and cancer .
