Background

Phospho-IκBα (Ser32) is a phosphorylated form of the IκBα protein, which plays a crucial role in the NF-κB signaling pathway. IκBα normally inhibits NF-κB by binding to it and preventing its nuclear translocation. However, when IκBα is phosphorylated at Ser32 (and often also at Ser36), it undergoes ubiquitination and subsequent degradation by the 26S proteasome, thereby releasing NF-κB to translocate to the nucleus and activate gene expression. This phosphorylation event is typically mediated by the IKK complex in response to various stimuli such as cytokines, growth factors, and stress signals. Antibodies specific to Phospho-IκBα (Ser32) are widely used in research to detect and quantify this phosphorylated protein, helping to elucidate the activation status of the NF-κB pathway in different cellular contexts.
