Background
DNA Ligase IV, also known as LIG4, is a crucial enzyme involved in the non-homologous end joining (NHEJ) pathway, which is essential for repairing DNA double-strand breaks. It plays a vital role in maintaining genomic stability by facilitating the ligation of broken DNA ends. LIG4 forms a complex with other proteins such as XRCC4 and XLF, enhancing its stability and activity. This enzyme is particularly important in the repair of DNA damage caused by ionizing radiation and certain chemotherapeutic agents. Mutations or deficiencies in LIG4 can lead to severe immunodeficiency and developmental disorders due to impaired V(D)J recombination, a process critical for the generation of antigen receptor diversity in the immune system.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
