Background

FUS (Fused in Sarcoma) is a multifunctional, predominantly nuclear DNA/RNA binding protein that plays a significant role in various cellular processes, including transcription, RNA splicing, and mRNA transport1. It belongs to the FET (FUS/EWS/TAF15) family of proteins and contains several functional domains that facilitate its interactions with nucleic acids, such as an RNA recognition motif (RRM) and a zinc finger domain. Mutations or abnormal expression of FUS have been implicated in several neurodegenerative diseases, most notably Amyotrophic Lateral Sclerosis (ALS) and Frontotemporal Lobar Degeneration (FTLD). In these diseases, FUS protein tends to form insoluble aggregates in neurons, which is thought to contribute to neuronal dysfunction and degeneration. The exact mechanisms by which FUS contributes to disease pathology are still under investigation, but it is believed that the protein's role in RNA metabolism and its mislocalization to the cytoplasm may be key factors.
