Background
TGN46 (Trans-Golgi Network protein 46 kDa) is a type I integral membrane protein predominantly localized to the trans-Golgi network (TGN), where it functions as a critical component of the protein sorting and vesicular trafficking machinery. This 46-kilodalton protein features a large N-terminal lumenal domain containing multiple N-glycosylation sites, a single transmembrane helix, and a short C-terminal cytoplasmic tail harboring sorting signals—including tyrosine-based motifs (YXXΦ)—that facilitate its interaction with adaptor protein complexes (AP-1 and AP-2) to regulate cargo selection and vesicle formation. TGN46 serves as a widely used marker for the TGN in immunofluorescence and cell biology studies, and plays essential roles in maintaining TGN structure, recycling mannose-6-phosphate receptors (MPRs) between endosomes and the Golgi, and sorting lysosomal enzymes into clathrin-coated vesicles destined for late endosomes. Its dynamic cycling between the TGN, endosomes, and the plasma membrane—mediated by phosphorylation-dependent conformational changes and interactions with various coat proteins—underscores its significance in secretory and endocytic pathway regulation, making it indispensable for proper cellular homeostasis and protein trafficking fidelity.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry
ICC shows positive staining in A549 cells. Anti-TGN46 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
