Background
Recombinant Trypsin (rTrypsin) is a genetically engineered serine protease (EC 3.4.21.4) produced via recombinant DNA technology, most commonly in microbial host systems such as Pichia pastoris or E. coli, to yield an enzyme that is identical in amino acid sequence to native porcine or bovine trypsin while ensuring an animal-free production process free from contaminants like chymotrypsin and risks of viral or transmissible spongiform encephalopathy (TSE) agents. As a key biocatalyst, it specifically hydrolyzes peptide bonds at the carboxyl termini of lysine and arginine residues, a specificity determined by its conserved catalytic triad (His57, Asp102, Ser195) and the substrate-binding S1 pocket, with its structural integrity maintained by a calcium-binding loop.

Linearity
The concentrations of Recombinant Trypsin (rTrypsin) were measured and interpolated from the target standard curves and corrected for sample dilution.
Sample is undiluted samples are as follows: 0.25 % Trypsin-EDTA (2.5 g/L trypsin, 0.38 g/L EDTA·4Na) was 6.25×104 fold dilution. The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 33.52 μg/mL in 0.25 % Trypsin-EDTA.
To assess the linearity of the assay, four samples were spiked with high concentrations of Recombinant Trypsin (rTrypsin) in various matrices and diluted with the appropriate Calibrator Diluent to produce samples with values within the dynamic range of the assay.
