Background

Sortilin 1 (SORT1) is a type I transmembrane glycoprotein encoded by the SORT1 gene, belonging to the vacuolar protein sorting 10 protein (Vps10p) family of sorting receptors. It is ubiquitously expressed in many tissues, with particularly high levels in the central nervous system. SORT1 plays a critical role in intracellular protein transport between the Golgi apparatus, endosomes, lysosomes, and plasma membrane, and is involved in various biological processes such as glucose and lipid metabolism, neural development, and cell death. The Vps10p domain of SORT1 forms a ten-bladed beta-propeller structure with multiple ligand binding sites, allowing it to bind over 50 proteins. SORT1 is also implicated in several diseases, including hypertension, atherosclerosis, coronary artery disease, Alzheimer’s disease, and cancer. Additionally, the SORT1 gene is located at a locus strongly associated with LDL cholesterol levels in genome-wide association studies.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-SORT1 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in Neuro-2a cells. Anti-SORT1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
