Background

CD22, or cluster of differentiation-22, is a molecule belonging to the SIGLEC family of lectins. It is found on the surface of mature B cells and to a lesser extent on some immature B cells. CD22 functions as an inhibitory receptor for B cell receptor (BCR) signaling. It is also involved in the B cell trafficking to Peyer's patches in mice. In mice, it has been shown that CD22 blockade restores homeostatic microglial phagocytosis in aging brains. Because CD22 is restricted to B cells, it is an excellent target for immunotherapy of B cell malignancies. There are several mechanisms by which this can be achieved, namely monoclonal antibodies, bispecific antibodies, antibody-drug conjugates, radioimmunoconjugates or CAR-T cells.

Flow cytometric analysis of Jurkat (Human T cell leukemia T lymphocyte, left) / Daudi (Human Burkitt's lymphoma lymphoblast, right) cells labelling Siglec-2 antibody at 1/200 dilution (1 μg) / (red) compared with a Mouse monoclonal IgG (Black) isotype control and an unlabelled control (cells without incubation with primary antibody and secondary antibody) (Blue). Negative control: Jurkat

Flow cytometric analysis of human PBMC (human peripheral blood mononuclear cell) labelling Siglec-2/CD22 antibody at 1/2000 (0.1 μg) dilution (Right) compared with a Mouse monoclonal IgG isotype control (Left). Goat Anti - Mouse IgG Alexa Fluor® 488 was used as the secondary antibody. Then cells were stained with CD19 - Brilliant Violet 421™ separately. Gated on total viable cells.

Immunocytochemistry

ICC shows positive staining in Daudi cells. Anti- Siglec-2/CD22 Mouse antibody was used at 1/250 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Mouse IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue).
