Background

Stress-activated protein kinases (SAPK)/c-Jun N-terminal kinases (JNK) are members of the mitogen-activated protein kinase (MAPK) family, activated by various environmental stresses such as UV irradiation, inflammatory cytokines, and growth factors. They play crucial roles in regulating cell proliferation, apoptosis, motility, metabolism, DNA repair, and immune responses. The JNK/SAPK pathway involves a three-tiered kinase cascade, with MAPK kinases (MKKs) like MKK4 and MKK7 phosphorylating and activating JNKs. This pathway is essential for cellular adaptation to stress and has been implicated in diseases such as cancer and metabolic disorders.

Immunocytochemistry

ICC shows positive staining in NIH/3T3 cells. Anti- SAPK/JNK antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
