Background

ROBO1 is a ~165 kDa single-pass transmembrane receptor of the immunoglobulin superfamily whose extracellular region contains five Ig-like domains and three fibronectin repeats that bind the secreted SLIT1/2 ligands, while its cytoplasmic tail carries four conserved CC motifs (CC0-CC3) that recruit adaptors such as srGAPs, Ena/VASP and DCC to transduce repulsive or attractive signals regulating axon guidance at the CNS midline, neuronal precursor migration, angiogenesis and even tumor metastasis by modulating Rho-family GTPases and actin cytoskeleton dynamics.

Immunocytochemistry

ICC shows positive staining in HeLa cells. Anti-ROBO1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
