Background
The RFX5 protein is a core subunit of a key transcription factor complex regulating adaptive immunity; its canonical function involves assembling with RFXAP and RFXANK into a heterotetramer (an RFX5 dimer bound to one RFXAP and one RFXANK molecule), recognizing the X-box motifs in MHC-I and MHC-II promoters via its N-terminal conserved domain, and utilizing its C-terminal domain to collaborate with NF-Y and other factors in stabilizing transcriptional enhanceosomes—deficiencies in this function lead to primary immunodeficiencies such as bare lymphocyte syndrome (BLS). The latest research revealed at the structural level that RFX5 also acts as a unique pioneer transcription factor: its extended DNA-binding domain (eDBD) directly forms hydrogen bonds with histones via the Glu102 residue, specifically targeting the SHL+2 position on nucleosomes. This induces localized DNA distortion and the dissociation of distal DNA from the nucleosome surface, thereby actively opening chromatin to enhance DNA accessibility and paving the way for subsequent transcription factor binding.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
