Background
Phospho-Stat6 (Tyr641) protein is a phosphorylated form of the Signal Transducer and Activator of Transcription 6 (Stat6) protein, specifically phosphorylated at tyrosine residue 641. Stat6 is a key transcription factor involved in mediating the cellular response to cytokines, particularly interleukin-4 (IL-4) and interleukin-13 (IL-13), which are crucial for the development and function of Th2 cells and the regulation of allergic responses and immune tolerance. When phosphorylated at Tyr641, Stat6 becomes activated and translocates to the nucleus, where it binds to specific DNA sequences to regulate the expression of genes involved in immune cell differentiation, cytokine production, and the development of allergic inflammation. This phosphorylation event is critical for the downstream signaling cascade initiated by IL-4 and IL-13 receptor activation, making phospho-Stat6 (Tyr641) a key biomarker for studying Th2-mediated immune responses and potential therapeutic target for allergic and immune-related diseases.

Immunocytochemistry
ICC analysis of Daudi cells starved for 24h then treated with IL-4 (100ng/ml, 15min) (top panel) and untreated Daudi cells (below panel). Anti- Phospho-STAT6 (Tyr641) antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
