Background

Phospho-FAK (Tyr397) is a crucial phosphorylated form of the focal adhesion kinase (FAK) protein, which plays a significant role in various cellular processes. FAK is a non-receptor protein tyrosine kinase that is involved in the regulation of cell adhesion, migration, and survival. When phosphorylated at tyrosine residue 397, FAK becomes activated and serves as a binding site for other signaling molecules, such as Src, thereby initiating downstream signaling pathways that are essential for cell motility and the formation of focal adhesions. Dysregulation of Phospho-FAK (Tyr397) has been implicated in several diseases, including cancer, where it contributes to tumor progression and metastasis by enhancing cell invasion and survival.

Immunocytochemistry

ICC analysis of HeLa cells treated with Pervanadate (10mM, 60min) (top panel) and untreated HeLa cells (below panel). Anti- Phospho-FAK (Tyr397) antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
