Background

NPHS2 protein, also known as podocin, is a key component of the slit diaphragm in the glomerular filtration barrier and plays a crucial role in regulating glomerular permeability; mutations in this protein can lead to steroid-resistant nephrotic syndrome.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human kidney. Anti-NPHS2 antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Negative control: IHC shows negative staining in paraffin-embedded human skeletal muscle. Anti-NPHS2 antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin.
