Background

Cleaved Caspase-8 (Asp387) is the ~18 kDa C-terminal catalytic fragment generated by autoproteolysis at Asp387 during death-receptor-mediated apoptosis, retaining the active-site cysteine and small subunit that, once released from the DISC and freed from its prodomain, forms a heterotetrameric protease capable of cleaving downstream caspases and substrates such as Bid, thereby amplifying the extrinsic apoptotic signaling cascade.

Immunocytochemistry

ICC analysis of RAW264.7 cells treated with Cycloheximide(2.5ug/ml,4h) and mTNF-α (20ng/ml,4h) (top panel) and untreated RAW264.7 cells (below panel). Anti- Cleaved Caspase-8 (Asp387) antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
