Background

ABCC1, also known as multidrug resistance-associated protein 1 (MRP1), is a 180–190 kDa integral membrane protein that belongs to the ATP-binding cassette (ABC) transporter superfamily. It contains two membrane-spanning domains, each with six α-helices, and two nucleotide binding domains, along with an additional third membrane-spanning domain. This protein functions as a multispecific organic anion transporter, with substrates including oxidized glutathione, cysteinyl leukotrienes, activated aflatoxin B1, glucuronides, and sulfate conjugates of steroid hormones and bile salts. It is highly conserved and expressed in various tissues, particularly in the lungs, spleen, testes, kidneys, placenta, thyroid, bladder, adrenal glands, and endothelium cells of the blood-brain barrier. ABCC1 plays a significant role in multidrug resistance of cancerous tumor cells by transporting chemotherapeutic drugs out of the cells. Additionally, it has been a point of interest in Alzheimer's disease research due to its ability to decrease β-amyloid accumulation in the brain.
