Background
Human Transforming Growth Factor-beta 1 (TGF-β1) is a highly conserved, disulfide-linked homodimeric cytokine (25 kDa) that plays crucial roles in cell proliferation, differentiation, and immune regulation. It is synthesized as a latent precursor consisting of a signal peptide, latency-associated peptide (LAP), and the mature TGF-β1 domain. The mature bioactive form is released from LAP through proteolytic cleavage or acid treatment (pH ≤ 3.0), which disrupts non-covalent interactions while preserving the dimer's structural integrity via critical disulfide bonds. Human TGF-β1 shares the following identity with other species: Monkey (Macaca spp.): ~99%; Mouse (Mus musculus): ~98%; Rat (Rattus norvegicus): ~98%; Rabbit (Oryctolagus cuniculus): ~97%; Donkey (Equus asinus): ~96%; Goat (Capra hircus): ~95%; Horse (Equus caballus): ~96%. The high homology underscores TGF-β1’s critical role in fundamental biological processes across mammals, enabling translational research using animal models. However, species-specific post-translational modifications or receptor interactions may still influence functional outcomes.

Sample is undiluted samples are as follows: human PBMC cells culture for 3days (50%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 1185.88 pg/mL in unstimulated human PBMC supernatant.
