Background

MUC1 is a transmembrane protein normally expressed in glandular or luminal epithelial cells and hematopoietic cells and absent in skin epithelium and mesenchymal cells. Apical surface of almost all glandular and ductal epithelial cells, including respiratory, gastrointestinal, urogenital, hepatobiliary tracts, sebaceous and salivary glands. MUC1 is expressed in the central cells of pancreatic acini, intercalary duct epithelium, and interlobular duct epithelium, but not in the acinar epithelium, main pancreatic duct epithelium, and pancreatic islet epithelium. Overexpression is common in malignancies and leads to tumorigenesis. Increased expression is associated with poor prognosis of most carcinomas, including those involving breast, cervical, pancreatic, colorectal.

FC

Flow cytometric analysis of 4% PFA fixed 90% methanol permeabilized HCT116 (Human colorectal carcinoma epithelial cell, left) / T-47D (Human ductal breast epithelial tumor epithelial cell, Right) labelling MUC1 antibody at 1/2000 dilution (0.1 μg) / (Red) compared with a Mouse monoclonal IgG (Black) isotype control and an unlabelled control (cells without incubation with primary antibody and secondary antibody) (Blue). Goat Anti - Mouse IgG Alexa Fluor® 488 was used as the secondary antibody.

Negative control: HCT116

IP

MUC1 Mouse mAb at 1/200 dilution (1 µg) immunoprecipitating MUC1 in 0.4 mg T-47D whole cell lysate.
Western blot was performed on the immunoprecipitate using MUC1 Mouse mAb at 1/1000 dilution.
Secondary antibody (HRP) for IP was used at 1/2000 dilution.
Lane 1: T-47D whole cell lysate 20 µg (Input)
Lane 2: MUC1 Mouse mAb IP in T-47D whole cell lysate
Lane 3: Mouse monoclonal IgG1 IP in T-47D whole cell lysate
Predicted MW: 122 kDa
Observed MW: 24 kDa
(This blot was developed with high sensitivity substrate)
