Background

Siglecs are transmembrane proteins of the immunoglobulin superfamily that bind sialic acid. They include an N-terminal V-set Ig-like domain and various C2-set domains. The first characterized Siglecs, including Siglec-1 to -4, show 25–30% sequence identity in their extracellular regions. A cluster of Siglec genes related to CD33 is found on human chromosome 19q13.3-4. The CD33-related subgroup (Siglecs-3, -5, -6, -7, and -8) shares 50–70% sequence identity. Each Siglec has a unique role in hemopoietic cell biology.
CD33 (Siglec-3), the smallest Siglec member, binds α2-6- and α2-3-sialylated glycans and is highly expressed on leukemic cell lines. It's primarily found on myeloid lineage leukocytes but also appears on lymphoid cells, including NK cells. CD33 exists as two membrane isoforms: full-length CD33M and CD33m, which lacks the V extracellular domain. CD33 expression is linked to Alzheimer’s disease, autoimmune conditions like SLE, type II diabetes, and infections.
