Instructions for use (for reference only)
· It is recommended to centrifuge at 100-200g for 2 minutes before use, then carefully open the lid and add 1mM HCl to reconstitute to a final concentration of 1mg/ml, then cover the tube and invert or shake to mix thoroughly. After reconstitution, it can be stored at 4℃ or -20℃ for at least 3 months. Try to avoid repeated freezing and thawing after reconstitution.
· After reconstitution, this product can be directly used for peptide mapping analysis or protein identification. The recommended addition ratio is 1:20-1:200, and enzymatic hydrolysis is carried out at 25℃ for 2-18h.

Background

Alpha-chymotrypsin, also known as alpha-chymotrypsin, is a serine protease that hydrolyzes the carboxy-terminal peptide bonds of tyrosine, tryptophan, and phenylalanine. Ca2+ is an activator and stabilizer of α-chymotrypsin, and the enzyme is supported by diisopropyl fluorophosphate (DFP), toluenesulfonyl fluoride (PMSF), n-p-toluenesulfonyl-l-phenylalanine chloromethyl ketone (TPCK), chymotrypsin inhibitors, aprotinin, α1-antitrypsin, α2-macroglobulin, and α-2-antitrypsin. Cu2+ and Hg2+ were inhibited by 10mM.
The recombinant human α-chymotrypsin produced by our company was expressed by Pichia pastoris and obtained by multi-step purification without contamination by other proteases.

Unit Definition

At pH 7.8 and 25°C, the amount of enzyme that hydrolyzes 1.0 μmol BTEE per minute is defined as 1 U.
