Background
pH630 Deep Red TFP Ester** is an amine-reactive dye that readily reacts with primary amines on biomolecules to form covalently linked fluorescent pH probes. This dye can be used to label primary amines on proteins, cells, or viruses, creating stable conjugates that fluoresce only in acidic environments. Our low-background pH630 Deep Red dye simplifies the study of internalization processes, yielding more reliable results with less optimization, as it is activated only in late endosomes and lysosomes. With excitation and emission maxima at approximately 640 nm and 655 nm, respectively, pH630 Deep Red can be detected using standard Cy5 filters.
pH630 Deep Red** is a low-background pH sensor dye that exhibits no signal under neutral conditions and fluoresces only in acidic environments. This unique property enables the rapid development and definitive results of assays for antibody internalization, endocytosis, and phagocytic pathways. The dye's approximate pKa value of 5 ensures activation only upon entry into late endosomes and lysosomes, allowing for better distinction between intracellular and extracellular materials. Compatible with Cy5 fluorescence filter sets, pH630 Deep Red has been validated for use in a variety of applications, including flow cytometry, fluorescence microscopy, and high-content screening (HCS).
