Background

ADAM22 is a member of the ADAM (A Disintegrin And Metalloprotease) family of transmembrane proteins. Unlike many family members, it lacks proteolytic activity and functions primarily as a scaffold protein within the central nervous system. Its key role is to anchor the protein complex at the synapse that includes postsynaptic receptors and signaling molecules. By interacting with specific partners like LGI1 and PSD-95, ADAM22 is crucial for organizing the synaptic architecture at excitatory synapses, thereby facilitating proper neuronal communication. Mutations in genes encoding ADAM22 or its binding partners are linked to severe epileptic disorders.
