Background
CD73, or ecto-5'-nucleotidase, is an enzyme encoded by the NT5E gene, forming a homodimer of two 70-kD subunits anchored to the plasma membrane's exterior. Its principal function is the conversion of extracellular AMP to adenosine, a key signaling molecule, distinguishing it from intracellular 5'-nucleotidases. Clinically, CD73 is a vital marker for lymphocyte differentiation, and its deficiency is linked to various immunodeficiency diseases, highlighting its role in immune function. Furthermore, rare mutations in the NT5E gene cause a distinct pathology known as adult-onset calcification of joints and arteries (CALJA). This syndrome involves calcification of the iliac, femoral, and tibial arteries, leading to impaired circulation in the legs and pain in the joints of the hands and feet. Therefore, CD73 is critically positioned at the intersection of purinergic signaling and human disease, with implications spanning from its fundamental role in generating adenosine to its clinical significance in immune deficiencies and vascular calcification disorders, making it a relevant biomarker and potential therapeutic target.
