Background

VDAC2 (Voltage-Dependent Anion Channel 2) is an evolutionarily conserved, ~32 kDa integral β-barrel porin embedded in the outer mitochondrial membrane where it orchestrates the flux of ATP, ADP, metabolites, and ions between the cytosol and mitochondria, serves as a crucial docking platform for pro- and anti-apoptotic Bcl-2 family proteins to modulate cytochrome-c release and intrinsic apoptosis, interacts with hexokinases to couple glycolysis to oxidative phosphorylation in cancer metabolism, and responds to transmembrane potential and pharmacological effectors such as erastin in ferroptosis; its expression is nearly ubiquitous yet highest in heart, skeletal muscle, and brain, and dysregulation contributes to neurodegeneration, cardiomyopathy, and chemoresistance, making VDAC2 a convergent therapeutic target for metabolic reprogramming and cell-death modulation.
