Background
TSG101 (Tumor Susceptibility Gene 101) is an essential 380-amino-acid human protein that contains an N-terminal UEV (ubiquitin E2 variant) domain and a proline-rich C-terminal region, functioning as the core subunit of the ESCRT-I complex that drives membrane scission events during multivesicular body formation, viral budding and cytokinesis by recognizing mono-ubiquitinated cargo via its UEV domain and coupling it to downstream ESCRT machinery, while its misregulation or truncation, originally identified in a screen for tumor suppressor genes, is linked to neoplastic transformation, defective cell cycle checkpoints and chromosomal instability.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
