Background

Stat5b (signal transducer and activator of transcription 5b) is a 787-amino-acid transcription factor that, upon phosphorylation by JAK kinases in response to cytokines such as IL-2, GH, prolactin or IFN-α, dimerizes and translocates to the nucleus where it binds the consensus GAS motif TTCNNNGAA to regulate genes controlling cell survival, proliferation, differentiation and metabolic reprogramming; it is indispensable for normal immune homeostasis, mammary gland development and liver GH signaling, and its persistent activation is a driver of multiple hematologic and solid cancers, making Stat5b a validated therapeutic target.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human colon. Anti-stat5b antibody was used at 1/2000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in RAW264.7 cells. Anti-Stat5b antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
