Background

Sirt3, a member of the sirtuin family of proteins, is an important enzyme that is primarily located in the mitochondria. It functions as a deacetylase, removing acetyl groups from various proteins, which helps regulate their activity and stability. Sirt3 plays a crucial role in cellular metabolism, stress resistance, and aging processes. By modulating the activity of key metabolic enzymes, it helps maintain mitochondrial function and energy homeostasis. Additionally, Sirt3 is involved in protecting cells against oxidative stress by influencing the expression and activity of antioxidant enzymes. Its dysregulation has been implicated in various diseases, including metabolic disorders and cancer, making it a potential target for therapeutic interventions.

Immunocytochemistry

ICC shows positive staining in HaCAT cells. Anti-Sirt3 antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
