Background
SIDT2 (Systemic RNA Interference Defective Transmembrane Protein 2) is a highly conserved transmembrane protein primarily localized to the lysosomal membrane in mammals, where it functions as a critical regulator of nucleic acid transport and lysosomal homeostasis. Originally identified through homology with the C. elegans SID-1 protein involved in systemic RNA interference, mammalian SIDT2 has evolved distinct roles centered on facilitating the uptake of extracellular double-stranded RNA (dsRNA) and mediating the degradation of intracellular RNA within lysosomes, thereby influencing gene silencing pathways and immune responses. Beyond its involvement in RNA metabolism, SIDT2 plays a significant part in autophagic flux by promoting the fusion of autophagosomes with lysosomes and assisting in the clearance of protein aggregates, which is particularly vital for neuronal health; consequently, dysregulation or mutations in the SIDT2 gene have been linked to neurodegenerative conditions, metabolic disorders, and altered susceptibility to viral infections, highlighting its multifaceted importance in maintaining cellular stability and managing nucleic acid trafficking across biological barriers.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-SIDT2 antibody was used at 1/250 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
