Background

RB1CC1 (also known as FIP200) is a multifunctional 183-kDa scaffolding protein that nucleates the ULK1-ATG13-RB1CC1-ATG101 complex to initiate autophagosome formation in response to nutrient or mTOR signals, acts in the nucleus as a GC-rich DNA-binding transcription factor that up-regulates RB1, p16 and p21 to enforce cell-cycle checkpoints, and simultaneously controls migration, differentiation and survival by inhibiting the focal-adhesion kinases PTK2/FAK1 and PTK2B/PYK2 and by modulating TSC1, p53 and TGF-β pathways, thereby operating as a central tumor-suppressor whose loss or mis-localation is linked to aggressive breast, prostate and pancreatic cancers as well as to fibrotic and neuro-developmental disorders.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human kidney. Anti-RB1CC1 antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
