Background

Rab11 is a member of the Rab family of small GTPases that plays a pivotal role in membrane trafficking, particularly in the recycling of membrane components from endosomes back to the plasma membrane. It is predominantly found in recycling endosomes and is crucial for the transport of vesicles from these endosomes to the cell surface, facilitating the reuse of receptors and membrane proteins. Its function has been well-defined in the recycling pathway, but its role in early endosomes is less understood. Recent studies have also implicated Rab11 in the balance of the endosome-lysosome pathway, suggesting its broader impact on cellular homeostasis. Overall, Rab11 is a key regulator in the maintenance of cellular membrane dynamics and the control of intracellular trafficking routes.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-Rab11 antibody was used at 1/250 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in A549 cells. Anti-Rab11 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
