Background

PSME4 (Proteasome Activator Subunit 4), also known as PA200 or Blm10 in yeast, is a nuclear proteasome activator that uniquely stimulates the peptidase activity of the 20S core particle through a gate-opening mechanism independent of ATP and ubiquitin conjugation. Structurally, PSME4 contains an HbYX (hydrophobic-tyrosine-X) motif at its C-terminus that docks into the inter-subunit pockets of the 20S α-ring, inducing conformational changes that facilitate substrate entry into the catalytic chamber. Unlike the ATP-dependent 19S regulatory particle or the PA28/11S REG family, PSME4 forms a dome-shaped homodimer that caps the 20S proteasome and preferentially promotes the degradation of unstructured peptides and certain natively disordered proteins, including acetylated histones and unstructured regions of prion-like proteins. PSME4 plays critical roles in spermatogenesis, where it is highly expressed in testis and essential for histone-to-protamine exchange during sperm chromatin remodeling, as well as in DNA damage response and cellular stress adaptation. Dysregulation of PSME4 has been implicated in various pathological conditions, including male infertility, neurodegenerative disorders characterized by protein aggregation, and cancer, where its expression levels correlate with tumor progression and therapeutic resistance in certain malignancies.

Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
