Background
Progranulin is a multifunctional protein that plays crucial roles in various biological processes. It is highly expressed in numerous tissues and is involved in cell growth, differentiation, and survival. Progranulin acts as a growth factor, promoting the proliferation of epithelial cells and fibroblasts. It also has anti-inflammatory properties, modulating immune responses by inhibiting the production of pro-inflammatory cytokines. Additionally, progranulin is implicated in the maintenance of tissue homeostasis and repair, particularly in the brain and nervous system. Mutations in the progranulin gene have been linked to neurodegenerative diseases such as frontotemporal dementia, highlighting its importance in neuronal health.

IHC shows positive staining in paraffin-embedded human spleen. Anti-Progranulin antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
